About Me
I am Dr. Yogeshwari Singh, a scientific researcher and consultant with a Ph.D. in Biological Sciences, an MBA, and 8+ years of research experience. My work spans molecular biology, protein biochemistry, RNA biology, structural biology, and biophysical characterization, with experience designing and executing independent research projects and developing and optimizing biochemical and molecular assays.
My technical expertise includes protein and RNA characterization, protein purification, molecular biology, RNA analysis, Northern blotting, enzyme kinetics, FPLC/SEC, SDS-PAGE, surface plasmon resonance (SPR), bio-layer interferometry (BLI), microscopy, structural biology, X-ray crystallography, and protein–protein and protein–ligand interaction assays. I also have experience analyzing complex scientific data, reviewing scientific literature, troubleshooting experimental challenges, and developing reproducible research workflows.

Molecular & RNA Biology
I work across molecular biology and RNA biology, with experience in molecular cloning, PCR, primer design, plasmid construction, targeted mutagenesis, in vitro transcription (IVT), radiolabeled RNA, RNA characterization, Northern blotting, and biochemical RNA assays.
Protein Biochemistry & Characterization
My experience includes recombinant protein expression and purification, FPLC/SEC, SDS-PAGE, enzyme kinetics, biochemical assay development and optimization, protein characterization, and analysis of protein–protein and protein–ligand interactions.
Biophysical, Structural & Cell Biology
I have experience using SPR, BLI, and MST for biophysical characterization, along with X-ray crystallography and structural biology to investigate molecular interactions and mechanisms. Projects
NAD-RNA Decapping & RNA Biology
My doctoral research focused on understanding NAD-RNA decapping and its regulation by the E. coli enzymes NudC and RppH and the signaling molecule ppGpp. I investigated how these enzymes recognize and process NAD⁺-capped RNA and how ppGpp influences their decapping activity.
My work combined molecular biology, RNA biochemistry, and biophysical approaches, including in vitro transcription, radiolabeled RNA substrates, RNA decapping assays, Northern blotting, SPR, BLI, and enzyme kinetics. I also used X-ray crystallography and structural analysis to investigate protein–ligand interactions and the molecular basis of NudC function.
This research resulted in a published method for measuring enzymatic RNA decapping activity and contributed to additional manuscripts in preparation on ppGpp-regulated NAD-RNA decapping and the cellular impact of NudC and RppH through integrated transcriptomic and proteomic analysis.
Protein, Biophysical & Cell Biology
My research experience spans protein expression, purification, characterization, and biomolecular interaction analysis. I have worked with FPLC/SEC, SDS-PAGE, enzyme kinetics, biochemical assay development and optimization, and protein–protein and protein–ligand interaction assays, using SPR, BLI, and MST for quantitative biophysical characterization.
I have also applied structural biology approaches, including X-ray crystallography, to investigate protein structure and molecular interactions. My work contributed to research on the E. coli DEAD-box ATPase CsdA as an NAD⁺-capped RNA binding protein and its interaction with the NAD-RNA decapping enzyme NudC.
Beyond purified protein systems, my experience includes cell culture and translational research at Nemours, working with iPSC-based mono- and co-culture systems and cancer patient-derived cells. I have also used fluorescence microscopy to investigate protein localization and cellular processes, providing complementary cellular and molecular perspectives.

Developed and described a gel electrophoresis-based assay for measuring enzymatic RNA decapping activity, providing a straightforward approach to evaluate RNA decapping reactions and enzyme activity. This work established a reproducible method for studying RNA 5′-end processing and decapping.
The E. coli DEAD Box ATPase CsdA Is a NAD+ Capped RNA Binding Protein
Contributed to research demonstrating that the E. coli DEAD-box ATPase CsdA binds NAD⁺-capped RNA, providing insights into the recognition and regulation of NAD⁺-capped transcripts in bacterial RNA biology.
ppGpp Regulates NAD-RNA Decapping by E. coli NudC and RppH
The regulation of NAD-RNA decapping by the E. coli enzymes NudC and RppH. This work investigates how ppGpp modulates NAD-RNA decapping activity and provides insight into the connection between RNA processing and bacterial stress signaling. (Manuscript in preparation)

An integrated transcriptomic and proteomic analysis of the cellular impact of NAD-RNA decapping by NudC and RppH. This work combines RNA-seq and quantitative proteomics to investigate how loss of NudC and RppH affects bacterial gene expression and cellular pathways.(Manuscript in preparation)
